Protein and Glycoprotein Characterisation by Mass Spectrometry

چکیده

Proteins are polymers made up of amino acids linked via their alpha amino and carboxyl groups respectively, creating planar interconnecting amide bonds. The order of the twenty naturally occurring amino acids in the polymer is called the amino acid sequence or primary structure, and all other levels of protein structure, including secondary (alpha helix, pleated sheet or random coil), tertiary (three dimensional) structure, or quaternary structure (interaction with a ligand or subunit in space) are fundamentally determined by the unique amino acid sequence of the protein. Determining the sequence of an unknown protein has therefore been the holy grail of protein chemistry ever since Fred Sanger’s pioneering work on the structure elucidation of Insulin – the first protein sequence determination in the 1950s1.

Applications of mass spectrometry in proteomics.

Mass spectrometry has arguably become the core technology in proteomics. The application of mass spectrometry based techniques for the qualitative and quantitative analysis of global proteome samples derived from complex mixtures has had a big impact in the understanding of cellular function. Here, we give a brief introduction to principles of mass spectrometry and instrumentation currently use...

Proteome analysis of Cryptosporidium parvum and C. hominis using two-dimentional electrophoresis, image analysis and tandem mass spectrometry

Until recently, Cryptosporidium was thought to be a single species genus. Molecular studies now showthat there are at least 10 valid species of this parasite. Among them, two morphologically identical species, C.hominis and C. parvum are the most pathogenic identified to date and share 97% of identical genomes.Post-genomic analyses is therefore necessary to explore further the...

Characterisation of an intrinsically disordered protein complex of Swi5-Sfr1 by ion mobility mass spectrometry and small-angle X-ray scattering.

It is now recognized that intrinsically disordered proteins (IDPs) play important roles as hubs in intracellular networks, and their structural characterisation is of significance. However, due to their highly dynamic features, it is challenging to investigate the structures of IDPs solely by conventional methods. In the present study, we demonstrate a novel method to characterise protein compl...

Protein profiling and analysis of drug sensitive and multidrug resistant isolates of Mycobacterium tuberculosis by native polyacrylamide gel electrophoresis and mass spectrometry

Introduction: Tuberculosis (TB) remains a deadly infectious disease despite all the efforts to reduce its incidence. Spread of multidrug resistant TB has seriously undermined the efforts to control the disease globally. In this study protein expression profile of MDR and sensitive isolates of MTB were analyzed and compared in order to identify proteins, which could be used in prevention, diagno...
